Enzymes of the Glyoxylate Cycle in Germinating From the Plant Peanuts and Castor Beans

چکیده
of conversion of fat to carbohydrate in germinating castor beans. Of the four requisite enzymes, isocitritase (Equation 3) has been demonstrated by direct assay (l), while the presence of condensing enzyme (Equation 1), aconitase (Equation 2), and malate synthetase activity (Equation 4) has been inferred from isotope incorporation data. The rates of condensing enzyme and malate synthetase, as estimated from the isotope data, were considerably lower than the rate of fat conversion by the intact seed. More recently, a study with germinating peanut and sunflower seeds (3) yielded results similar to those obtained earlier with castor beans. In a previous communication from this laboratory (4), direct evidence was presented for the malate synthetase reaction in germinating peanuts, and a distribution study indicated that this reaction was associated primarily with high fat seeds. The present report is concerned with the rates of reaction of the various enzymes, their cellular localization, and the relationship to the germination process. Data on TPN-isocitric dehydrogenase are also presented because of its relevance to competition for isocitrate by the glyoxylate and tricarboxylic acid cycles.

متن کاملOccurrence of RNA in glyoxysomes from castor bean endosperm.
The finding that in the endosperm of germinating castor bean, the enzymes of the glyoxylate cycle were housed in a special organelle, the glyoxysome (1, 2) raised the question of its biogenesis. We have shown that glyoxysome protein increases strikingly during the early stages of germination and concluded that rapid formation of new organelles occurs at this time (8). When the individual organe...
متن کاملIsoenzymes of sugar phosphate metabolism in endosperm of germinating castor beans.
Two isoenzymes each of phosphoglucomutase, hexose phosphate isomerase, aldolase, fructose diphosphatase, phosphofructokinase, and 6-phosphogluconate dehydrogenase have been separated by DEAE-cellulose column chromatography of extracts from endosperm of germinating castor beans (Ricinus communis cv. Hale). One of each of the enzymes is localized in the cytosol and the other is confined to plasti...
متن کاملThe enzymatic characteristics of peroxisomes of amphibian and avian liver and kidney.
Peroxisomes have been identified on the basis of morphological criteria in a variety of species (Hruban and Rechcigl, 1969). Biochemical characterization, however, has been limited to rat liver (deDuve & Baudhuin, 1966; Leighton, et al., 1968), rat kidney (Allen et al., 1965; deDuve & Baudhuin, 1966), Tetrahymena pyriformis (Baudhuin et al., 1965b; Miiller et al., 1968), Acanrhoamoeba (Miiller ...
متن کاملThe presence of ribulose 1,5-diphosphate carboxylase in the nonphotosynthetic endosperm of germinating castor beans.
Ribulose 1,5-diphosphate carboxylase was detected in extracts of germinating castor bean (Ricinus communis var. Hale) endosperms. This is the first report of this enzyme in a nonphotosynthetic (no chlorophyll) plant tissue. Radioactive 3-phosphoglyceric acid has been identified as the principle product resulting from the enzymatic condensation of (14)C-bicarbonate and ribulose-1,5-diP in endosp...
